I am writing to you today to share an exciting opportunity to develop a proposal for the Advanced Research Projects Agency for Health (ARPA-H) Computational ADME Tox AnaLYsis for Safer Therapeutics (CATALYST) program. This program has released an Innovative Solutions Opening Solicitation with a focus on developing AI/ML-enabled in silico human physiology modeling platforms for ADME-Tox simulation to replace the poorly predictive investigational new drug (IND) IND-enabling preclinical animal studies currently used.
This initiative aligns perfectly with our institute's mission to promote successful longevity and could significantly impact the field of drug discovery for Alzheimer’s disease and related dementia (and co-occurring conditions related to ADRD). The goal of the CATALYST program is to reduce drug development costs and timelines while increasing the success rate of clinical trials through accurate prediction of the safety, toxicity, and availability of therapeutic candidates.
The program is structured in two phases:
Phase I: Accelerated Technology Development and Method Qualification (36 months): Focuses on developing technologies that meet the in silico predictive goals of CATALYST through three technical areas (TAs):
○ TA1: Data discovery methods and predictive drug safety models4
○ TA2: Living systems tools for model development4
○ TA3: In silico simulation-based human physiology models4
Phase II: COU Concept Demonstration and Methodology Adoption (18 months): Drug developers pursue regulatory filings using the in silico platform developed in Phase I.
To ensure the success of this endeavor, we need a strong team with expertise in various areas, including data discovery, living systems tools, in silico modeling, and drug development. I am reaching out to gauge your interest in joining a discussion about developing a proposal for this opportunity.
I am particularly interested in hearing from faculty affiliates with expertise in ADRD and those with a strong background in AI/ML and computational modeling. The sources mention a few specific areas of expertise:

● Proteomics5
● Toxicological data
● Systems biology, including physiological data
● Preclinical study
● Clinical study and data (EHR)
